    A. Basic Cell Culture Techniques:

    Types of Cell Line Handled.
    Primary Cultures:
    I.Primary Culture of Dentritic Cells: Primary cultures of stem cells isolated from Bone marrow
    preparation are maintained and studied their propagation into dentritic cells.
    II.From Spleenocytes preparation: Purification and propagation of T-cell.
    Secondary Culture:
    Cancerous cell lines: Handling and maintenances of COLO and MIAPPACA cell line.
    B. FAC`S: Harvesting, Cell staining with Fluorescence Labeled Monoclonal Antibodies:
    Primary dendritic cells were treated with various phytochemicals, peptides, compounds of marine
    origin for the screening of their potential as the Immunomodulatory and Anti- inflammatory
    properties.
    • Some cells were harvested from the primary culture which is not treated with any chemical
    compound, stained with fluorescent dye attached to monoclonal antibodies; cells were then subjected
    to Flow Cytometry (fluorescence activated cell sorter) which analyses large population of cell for the
    amount of receptor on their surface as the level of fluorescence.
    • Some cells from the primary culture were subjected to various chemical treatments
    periodically, then stained with fluorescent dye attached to monoclonal antibodies and then subjected
    to Flow Cytometry.
    • Preliminary known to data acquisition and data analysis using Cell Quest software.
    • Machine Used: FAC`S calibrator with four filters.
    C. Enzyme Linked Immunosorbent Assay (ELISA)
    • Used commercially available kits for detecting cytokines examples, IL-6, IL-10, IL-12,
    RANTES, IFN-γ, TGF-β, and TNF-α .
    • These cytokines were secreted by cell population when exposed to various chemical
    compounds.
    • Through the analysis of results obtained from Elisa, it is calculated which chemical is
    responsible for the secretion of which cytokine, in this way surface reactor which facilitates the
    secretion can be traced.
    • Reference curves generated with linear regression curves fitting with r2 value >0.98.
    • Hands on experience of using Graph pad prism for data analyses and interpretation of result.
    D. Cytotoxic Assay (MTT Assay): MTT is yellow water soluble tetrazolium dye that is reduced by live
    cells to a purple formazen product that is insoluble in aqueous solution. This assay is performed to
    study the cytotoxicity of the drugs.
    IN VIVO Techniques:
    A. Identification and handling of small experimental animals like Rat and Mice.
    B. Administration of Drugs / cancerous cell lines in experimental animals through various routes
    of administration.

    • Lymph nodes for the study of migration of fluorescence labeled cells injected through
    Sub-cutaneous route.
    D. Necropsy performed on experimental animals which includes following steps;
    • Exsanguination of animal.
    • Gross examination of internal anatomy of animal.
    • Collection and preservation of various organs and glands for pathological examination.
    E. Blood with-drawl by capillary method for hematological analysis of experimental
     animals depending upon their body weight.
    F. Migration assay of C.F.S.E labeled T-cells in lymph nodes of lived mice.
    G. Study of various type tumor in nude mouse, migration of malignant cells in a living model.
